[editar] Biografía
Rupert nació en Londres Inglaterra, es hijo del actor Peter Penry-Jones y la actriz Angela Thorne; su hermano Laurence Penry-Jones también es actor. Rupert mide 6' 2" (1.88 m) y se educó en el Dulwich College, al sureste de Londres. En 1995 paticipó en la serie de televisión Cold Confort Granja, en donde compartió créditos con su madre. Su apodo es "Roo".
En 1999 salío con la cantante australiana Kylie Minogue durante 9 meses.
En el 2001 conoció a la actriz Dervla Kirwan en una producción de teatro y en el 2005 participaron en la película Casanova, aunque no compartieron ninguna escena juntos. En agosto de 2007, después de tres años de compromiso se casó con Kirwan, el primero de mayo del 2004 le dieron la bienvenida a su primera hija Florence y el 8 de abril del 2006 a su segundo hijo Peter.

Rupert entrenó en el Bristol Old Vic Theatre School.
En 1995 hizo su debut en Londres junto a Ralph Fiennes en una producción de Almeida acerca de Hamlet. En 1996 interpreto a Richard en Sweet Panic en una producción de Stephen Poliakoff en el Teatro Hampstead.
Al año siguiente apareció en las puestas en escena de The Paper Husband y Chips With Everything. En 1998 hizo el papel del niño en The Play About the Baby en el teatro Almeida. En 1999 le dio vida a Don Carlos en The Other Place y a Alcibiades en Timon of Athens en el Royal Shakespeare Theatre. Ambas producciones fueron transferidas al Barbican Centre en el 2000, su interpretación como Don Carlos le hizo merecedor al premio Ian Charleson.
Entre julio y octubre del 2003, participó en el Teatro Nacional de Cottesloe donde interpretaba a Louis XIV, en el drama histórico Power.
Más tarde en el 2003 se unió a la tercera temporada de Spooks, donde interpretó el papel de Adam Carter un agente encantador pero despiadado que fue transferido del MI6 para investigar la desaparición de Tom Quinn interpretado por Matthew Macfadyen y que luego tomaría el puesto del líder en el MI5; luego de que en el primer episodio de la séptima temporada en el 2007 su personaje mueriera en una exploción.
En el 2009 se unió al elenco de la serie drámatica de tres partes Whitechapel, interpretando al Detective Joseph Chandler; Whitechapel se basa en el asesinato de Jack el destripador. Ese mismo año también apareció en la serie The Forgotten donde interpretó a Alex Donovan
